THE NRLP3 INFLAMASOME IS ACTIVATED IN TYPE II PNEMOCYTES IN RESPONSE TO THE INFLUENZA VIRUS

Introdução

The NLRP3 inflammasome promotes inflammation through the cleavage and activation of inflammatory cytokines such as IL-1β and IL-18. NLRP3 activation can occur from endosomal TLR receptors, through viral RNA recognition. NLRP3 inflammasome activation is observed in influenza infection, contributing to exacerbated inflammation and adverse clinical outcomes, however, unresolved questions remain regarding the relationship between inflammasome activation and lung inflammation during this viral infection.

Objetivo (s)

To evaluate the role of NLRP3 inflammasome activation in Influenza A infection.

Material e Métodos

Wild-type C57Bl/6 mice and knock-out mice for Nlrp3, C1/11 and Asc/C1/11 (CEUA: 046/2020), and infected intranasally with Influenza A (CA09), using 10 4 PFU. The qPCR and ELISA assays were used to respectively quantify the viral load and IL-1β cytokine, as well as an immunofluorescence assay to count NLRP3 puncta. Finally, a total of 2x10 5 type 2 pneumocytes (A549 cells) were infected and infected with Influenza A at an MOI of 0.1 and 0.01.

Resultados e Conclusão

It was observed that the weight variation is similar in both groups, and 30% of the infected knock-out mice (Nlrp3-/-) survived the infection. We observed a lower viral load and greater release of IL- 1β in the Nlrp3-/- group. Regarding the NLRP3 puncta count, the activation of this inflammasome was observed both in the lungs of infected mice, and specifically in type 2 pneumocytes. In the in vivo analysis, the activation of NLRP3 is well distributed in the lung parenchyma, while in the in vitro analysis, the higher MIO, led to greater activation of the inflammasome. The NRLP3 inflammasome appears to contribute to an efficient response against infection.
